Pamilyang Pinoy Henyo is an edition of the segment Pinoy Henyo that premiered on 25 June 2012 and continued until 19 January 2013. It is the second edition of the Pamilyang Pinoy Henyo series.

Thousands of families face each other in the daily elimination round of the competition. Then, they undergo a series of elimination rounds before being narrowed down in the semi-finals. The contestants are further narrowed down into four pairs who ultimately competed in the grand finals of the competition.

The daily round of this edition follows the regular format and rules of the original Pinoy Henyo. The pair who correctly guesses the word with the fastest time advances to the jackpot round. In the jackpot round, the winning pair must take turns to correctly guess any three of the ten words provided in under three minutes.

In the semi-final round and last elimination rounds, pairs are given a set of ten words from any of the categories. Players can guess as much words as they can within three minutes. Pairs can opt to pass, but they cannot return to those skipped words. Whichever pair guesses the most words with the fastest time wins the game.

The grand finals of the competition is the same as the previous qualifying rounds with a slight modification. For the first time, the game implemented a point system in the grand finals. Players can guess as much words as they can from the given ten words that came from any of the categories. However, each word is worth a certain point ranging from 1 to 3 points. The more difficult the word is, the higher point it is worth. The pair with the highest number of points wins the game. In case of a tie in the number of points, the time when the first word is guessed will be the deciding factor of the winner. Whichever pair guesses the first word the fastest ultimately wins the game.   

The family clans who competed in the grand finals were the Despabiladerases, the Santiagos, the Hinayons, and the Ventureros.

Siblings Trix Despabiladeras and Harvey Despabiladeras of Quezon City were the winners of the competition after guessing nine words within 2 minutes 24 seconds. They received a trophy, a brand-new Chevrolet Spark vehicle, and ₱300,000 in cash prize.

The Despabiladeras Family is dubbed as the "Pamilyang Perfect 10" for guessing all ten words in most of their games during the semi-finals and the last elimination rounds. In the grand finals, they almost got a perfect ten once again, but the judges refused to count the point when they guessed the word "Dumaguete City" instead of just "Dumaguete."
