Jackpot En Poy is a game segment based on the classic children's game rock–paper–scissors. Four players are paired to compete in the three-round segment that follows a knockout format. In the first round, the first pair plays against each other until one player wins three times. The next pair then plays against each other in the second round. The winners from the first two rounds then compete against each other to determine the ultimate winner. The winner of the game then moves on to the final round. In the final round, the player is presented with several Dabarkads, each holding different amounts of cash prize. The player will then pick three Dabarkads who he or she will play rock–paper–scissors against. The player goes against them one at a time. If the player wins against any of the Eat Bulaga! host, he or she will win the cash prize.
The regular referee for the segment is Australian fitness trainer and lifestyle coach Gov Lloyd. Other guest celebrities who became the referee of the segment include Ruffa Gutierrez, Ai-Ai delas Alas, Brandon Vera, and Jay Contreras. Jose Manalo, Wally Bayola (as Lola Nidora), and Baste Granfon also became referees.
After the entertaining refereeing stint of Ruffa Gutierrez as the "sosyal na referee", Ruffa was invited back to Eat Bulaga! to become a regular referee in the segment Jackpot En Poy, earning the nickname "Ruffaree". Ruffa referees the game on Tuesdays and Thursdays, while Gov Lloyd referees the game the rest of the week.

- The 4 April 2017, 11 March 2017, and 25 March 2017 celebrity episodes were replayed in full on Monday, 27 April 2020 to Wednesday, 29 April 2020, respectively. Due to the COVID-19 pandemic that resulted in the suspension of all live shows, Eat Bulaga! opted to replay throwback videos of some of its most iconic segments and other special presentations from way back in the past.
- Interestingly, at least 2 out of 3 news reporters/anchors and personalities that played, all of which are from GMA Integrated News and GMA Public Affairs, were shutout (0–3). Ivan Mayrina and Arnold Clavio lost on separate episodes.
- The other GMA News personality, Nathaniel Cruz, won his episode and got a perfect 9 (9–3).
